Cancer Drug May Treat Alzheimer's (LiveScience.com)
A cancer drug has succeeded in reversing Alzheimer's disease in its early stages in mice, according to a new study.
The drug, bexarotene, is designed to reduce levels of amyloid beta, the protein whose presence in the brain has been most closely tied to the development of Alzheimer's.
In a new study, mice treated with bexarotene saw their amyloid beta levels drop 25 percent within six hours and, importantly, they showed a corresponding improvement in their cognitive function.
"The data we provide here really suggest that Alzheimer's could be, in the early stages, a reversible disease," said study author Paige Cramer, a doctoral student in neuroscience at Case Western Reserve School of Medicine.
The researchers used mice that had a mouse model of Alzheimer's disease. After the researchers administered varying doses of bexarotene, they measured levels of amyloid beta in the brain and tested the mice for their abilities in maze running, nest building, smell, and fear conditioning, which is a type of learning.
"They did a lot of different tests of learning and memory and they saw an effect on every single one of them," said Michael Sasner, a research scientist and associate director at the Jackson Laboratory in Bar Harbor, Maine.
Bexarotene is already approved by the Food and Drug Administration for the treatment of cutaneous T-cell lymphoma, a type of skin cancer, and so it may be able to proceed through clinical trials more quickly than drugs not already known to be safe to administer to people.
The study appears in the Feb.10 issue of the journal Science.
A new way to target Alzheimer's
Sasner, who was not involved with the new study, said it overcomes some of the weaknesses of previous Alzheimer's work, in which only one or two tests of cognitive improvement were conducted.
Bexarotene is not the first attempted Alzheimer's treatment to target amyloid beta. But past research has aimed at removing the plaques that amyloid beta can form in the brain, which has not shown any effect on the disease itself.
The difference now, researchers say, is in a better understanding of amyloid beta and the various forms it can exist in. Rather than focusing on the plaques, researchers now think it is the active, soluble form of the protein that is at work in Alzheimer's.
While there are various views on the causes of Alzheimer's, "the predominant view right now is that it's the soluble forms of amyloid beta that are causing the impaired brain function," Cramer said. "Plaques are just sinks, just tombstones that gather amyloid beta."
Bexarotene works by promoting the production of another protein, called Apolipoprotein E, which binds to and clears amyloid beta from the brain.
"This paper lends a lot to the mechanism of how ApoE may be involved in Alzheimer's," Cramer said.
Hurdles remain
It remains to be seen whether the benefits of bexarotene in mice would translate to humans.
"Because we're using an FDA-approved drug, this allows us to translate these basic science findings to the clinic; that's our next goal," Cramer said.
Figuring out the correct dosing presents another challenge. Researchers found that in one case, giving bexarotene over several doses appeared to be less effective than giving it once. Cramer said the reason may be that the drug degrades itself within the body.
Cramer said she and Gary Landreth, the senior investigator on the study, hope to begin a preliminary trial this year, in which they will look for the same changes in beta amyloid levels in humans. If successful, the testing would move to clinical trials.
Bexarotene is currently sold as Targretin; patents on that drug will expire in April.
"There's a long way to go to prove this treatment in humans, but it seems like an exciting thing to follow up on," Sasner said.

Country music disappears from Grammy spotlight (AP)
NASHVILLE, Tenn. ? Lady Antebellum landed an unexpected knockout of Eminem and took the night with five Grammys last year, including record and song of the year for "Need You Now." Taylor Swift and Zac Brown Band broke through in the general categories the year before.
Yet this year only one country nominee made the cut in the prestigious all-genre categories: new artist entry The Band Perry.
Some might suggest a country snub, but Jason Aldean, a first-time nominee who's up for three awards, including country album of the year, doesn't see this year's deficit as out of the ordinary. That the focus is on the likes of Adele, Bruno Mars and Rihanna is not surprising to him, even though he and Swift had two of the year's top-selling albums.
"All those artists are great and having a killer year, so it's hard to argue with what they're doing," Aldean said. "But I think it's something country music has fought for years and years. I don't think it's anything new. I don't think at least from my perspective it's something that I'd be shocked about. It's the way it always is. It's not going to change. It's just how it goes."
Aldean proves himself an astute student of country music history with that statement. The genre's ascendance in the last half of the last decade at the Grammys is an aberration, a statistical oddity based on the overwhelming crossover success of a handful of songs and artists who dominated pop music at the time of their wins.
Mainstream country has scored just 13 general-category trophies since the first Grammys in 1958. Just three of those came before 2000.
Lady A's win in the record of the year category in 2010 was just the second by a country artist. The Dixie Chicks, fresh from the band's excommunication by country radio after remarks about President George W. Bush, won three general category awards at the 2007 Grammys.
Aldean notes all those winners have something in common: crossover success. Especially Swift and Lady A.
"(Swift) was just as big in the pop world as she was in the country world," Aldean said. "That was just one of those you can't really deny. As an awards show you almost look stupid if she's not there. And I think Lady A last year, without a doubt they had one of the biggest songs, if not the biggest song, of the year. ... They had a huge year, a huge record. It's kind of hard to deny that."
Members of Lady A, up for country album of the year, said it didn't really matter in what categories their nominations fell. Hillary Scott, Dave Haywood and Charles Kelley are just happy to be back and able to spend time with good friends like Aldean, country song of the year nominee Dave Barnes, Eric Church and others.
"We'd be lying if we said we wouldn't be disappointed after such an incredible year there last year if we didn't get invited back to the party," Kelley said.
"It's fun to be able to support our friends in the industry, as well," Scott said. "There are a lot of our really close friends in the business who will be there that we can celebrate with and just enjoy the night."
Gary LeVox, lead singer for Rascal Flatts, thinks that's the right attitude to have. Rascal Flatts landed one of those coveted all-genre song of the year nominations for "Bless the Broken Road," but over time he's learned not to take it too seriously. It's in no way an exact science, after all.
"I know some Grammy voters that are actually friends of mine that only listen to country music, but they vote on categories in the rock world," LeVox said. "They don't know anything about the rock world and they'll tell you, `I don't know. I've just heard of Maroon 5 so I just voted for that.' And those are my friends. I think some of that happens."

Sikh Temple Graffiti Vandalism Leads To Call For Hate Crime ...
A Sikh temple or gurdwara under construction in Sterling Heights, Mich., was recently vandalized with graffiti, in what some are calling a hate crime.
The incident occurred late Sunday night or early Monday morning and is currently under investigation by the Sterling Heights Police Department. The graffiti on the building's outside wall depicts a gun and a Christian cross. Also legible are the words "Mohmed," potentially a reference to the Islamic prophet Mohammed, "Dont Builed," "fcuk you" and the letters "R" and "A."
Sterling Heights Police Public Information Officer Lt. Luke Riley said Wednesday nothing left at the site has helped identify the culprit. He said media attention to the case may cause a witness to step forward. Anyone with further information is asked to contact the department directly.
The graffiti's possible reference to Mohammed, the Islamic prophet who is not a figure in the Sikh religion, has caused some to argue the defacement was directed at Muslims, Michigan Radio reports.
The Sikh American Legal Defense and Education Fund (SALDEF) contacted local and state officials asking them to investigate the incident as a hate crime.
"We call upon local and federal law enforcement agencies to rightfully classify this incident as a hate crime and bring the perpetrators to justice to show that hate and violence are not tolerable in our society," SALDEF Associate Executive Director Jasjit Singh said in a statement.
The Michigan chapter of the Council on American-Islamic Relations (CAIR-MI) has also urged the incident be investigated as a hate crime, according to WXYZ.
The U.S. Department of Justice in Detroit has met with members of the Sikh congregation, but it's not clear how they might be involved in the investigation. SALDEF also contacted the office of the U.S. Attorney.
"Attacks and vandalism against any of the nation's houses of worship must be condemned by all Americans," said Singh.
According to WXYZ, The gurdwara is due to be finished in the summer and will serve 225 families.

The William Hall Library was the first official public library in Cranston when it opened as the Edgewood Library 1897 in a small house at the corner of Park and Warwick Avenues.
Hall, a prominent businessman, established a fund to form the library in his will. He died in 1916.
Hall (1837-1916) was an entrepreneur and a well-respected member of the Cranston community, serving on the Cranston City Council, state Senate and House of Representatives. He was never defeated in his numerous runs for office and legend has it he started his first business at 18 with just $50, selling fruits and vegetables from a small market on Broad Street.
The library was renamed in 1921 after the Edgewood Free Public Library was transitioned into the new William H. Hall Free Library, run by a board of trustees envisioned in Hall's will. After a lengthy public debate, a new, elegant building was built on the grounds of Hall's former mansion on Broad Street. It was opened to the public on Nov. 13, 1927.
The building combines elements of Georgian and Italian Renaissance design with a curved arches over the loggia and white pillars. Three double glass and bronze doors open to an impressive, high-cieling interior.?
The library boasts a large collection of fiction, nonfiction, reference and children's books. It also serves as a community center and gathering place.
Though he was childless, Hall was an ardent supporter of learning and children's issues. Long after his death, the William H. Hall Library stands as a monument to his desire to enrich the lives of Cranston's youth.
